چکیده

HLA-B*13:01 is associated with dapsone (DDS)-induced hypersensitivity and it has been shown that CD4+ CD8+ T cells are activated by DDS its nitroso metabolite (DDS-NO). However, there a need to define the importance of HLA association in disease pathogenesis. Thus, DDS-NO-specific cell clones (TCC) were generated from hypersensitive patients expressing assessed for phenotype function, HLA-allele restriction, killing target cells. TCC stimulated proliferate secrete effector molecules when exposed and/or DDS-NO. DDS-responsive several DDS-NO-responsive variety TCR sequences displayed class-I-restriction, drug(metabolite) interacting multiple HLA-B alleles. activation certain was inhibited HLA-class-II block, DDS-NO binding HLA-DQB1*05:01. These different origin, but expressed TCRs displaying same amino acid sequences. They via hapten pathway, CD45RO, CD28, PD-1 CTLA-4 surface molecules, secreted panel as class-I-restricted TCC, lower capacity lyse To conclude, interact number activate class-II-restricted display hybrid CD4/CD8 features also contributing promiscuous immune response develops patients.

HLA-DR-promiscuous T cell epitopes from Plasmodium falciparum pre-erythrocytic-stage antigens restricted by multiple HLA class II alleles.

Previously, we identified and established the antigenicity of 17 CD8+ T cell epitopes from five P. falciparum Ags that are restricted by multiple common HLA class I alleles. Here, we report the identification of 11 peptides from the same Ags, cicumsporozoite protein, sporozoite surface protein 2, exported protein-1, and liver-stage Ag-1, that bind between at least five and up to 11 different HL...

The Emerging Role of HLA-E-Restricted CD8+ T Lymphocytes in the Adaptive Immune Response to Pathogens and Tumors

Human leukocyte antigen (HLA)-E is a nonclassical major histocompatibility complex (MHC) class I molecule of limited sequence variability that is expressed by most tissues albeit at low levels. HLA-E has been first described as the ligand of CD94/NKG2 receptors expressed mainly by natural killer (NK) cells, thus confining its role to the regulation of NK-cell function. However, recent evidences...

Thymus dictates major histocompatibility complex (MHC) specificity and immune response gene phenotype of class II MHC-restricted T cells but not of class I MHC-restricted T cells

Athymic H-2b nude mice received grafts from C57BL/6 (Sendai virus and H-Y antigen cytotoxic T lymphocyte [CTL] responder type), bm1 (H-2Kb mutant, Sendai CTL nonresponder type), or bm12 (H-21-A mutant, H-Y CTL nonresponder type) neonates. In observations of the CTL response to H-Y, both recipients and thymus donors were female. All types of thymus engraftment resulted in mature H-2b splenic T l...

Irradiated β-glucan enhances immune response to bacterial infection through CD4 and CD8 T-lymphocytes

Background: &beta-glucans are glucose polymers with a variety of stimulatory effects on the immune system. The objective of this study was to evaluate the immune-enhancing activities of low molecular weight gamma irradiated &beta-glucan (I&beta-g) extracted from Pleurotus ostreatus in response to Pseudomonas aeruginosa infection in rats. Materials and Methods: &beta-glucan (&beta-g) powder was ...

The natural immune response to inhaled soluble protein antigens involves major histocompatibility complex (MHC) class I-restricted CD8+ T cell-mediated but MHC class II-restricted CD4+ T cell-dependent immune deviation resulting in selective suppression of immunoglobulin E production

The immunological basis for atopy is currently ascribed to an inherent bias in the CD4+ T cell response to nonreplicating antigens presented at mucosal surfaces, resulting in dominance of the T helper 2 (Th2) interleukin 4 (IL-4)-producing phenotype, which favors IgE production. In contrast, the "normal" response to such antigens involves a predominance of interferon gamma (IFN-gamma)-producing...
